Description
Microbiology and staining course using hands-on exercises to promote the use of the microscope for process control and troubleshooting of the activated sludge process. Major components of the mixed liquor examination that are reviewed include: 1) bulk solution, 2) floc particles, 3) filamentous organisms, 4) protozoa and 5) metazoa. Filamentous organism topics that are reviewed include the relative abundance of filamentous organisms and identification of filamentous organisms. A detailed review of the use of protozoa as “bio-indicators” of the “health” of the activated sludge and the quality of the mixed liquor effluent is provided.
Exercises performed include the preparation of wet mounts, stained wet mounts, smears and stained smears. Calibration of objective lenses for microscopic measurements is also performed.
